Decreased expression levels of BDNF and TrkB in the offspring rats’ hippocampus and the modulatory effects of DHF treatment. <t>Immunohistochemistry</t> staining revealed that the protein levels of BDNF and TrkB in the propofol exposure group (P group) were significant less than those in the control group (C group) ( A – C ). TrkB agonist (7,8‐ DHF ) reversed the expression of BDNF and TrkB. The protein levels of BDNF and TrkB in the 7,8‐ DHF treatment group ( PD group) were higher than those in the P group ( A – C ). Further, 7,8‐ DHF did not change the protein levels of BDNF and TrkB in rat offspring that had not been exposed to propofol ( CD group) ( A – C ). The changes of BDNF and TrkB mRNA expressions were similar to their proteins ( D ). ( A ) BDNF protein immunohistochemistry stain in offspring's hippocampus (400×); ( B ) TrkB protein immunohistochemistry stain in offspring's hippocampus (400×); ( C ) Data represents mean ± S.E. of 20 offspring rats in each group. ( D ) Real time PCR results of BDNF and TrkB mRNA expressions. Data represents mean ± S.E. of 20 offspring rats in each group, * P < 0.05.
